چکیده

A survey of chromosome patterns in human leukemia was aimed at determining: departures from diploidy in different types of acute and chronic leukemia, influence of therapy on chromosome constitution, frequency of aneuploidy in freshly aspirated marrow compared with the same sample after short-term tissue culture. Exact chromosome counts were obtained in over 3,600 metaphases from 34 leukemic (including two mongols) and 60 nonleukemic marrows. Human leukemias as a whole were characterized by a greater variability in chromosome structure and number (56.4 __+ 1.2 per cent aneuploid) than control marrows (12.2 + 0.8 per cent aneuploid). This was true even for leukemias with diploid modal karyotypes. Acute lymphoblastic leukemias as a group had the highest incidence of aneuploidy (77.0 + 1.6 per cent). Chemotherapeutic treatment did not seem to enhance chromosome anomalies; on the contrary, two aneuploid leukemias in remission during therapy reverted to normal counts of 46. Marrow samples from nine leukemic patients were examined by two methods: fixation immediately after sternal aspiration (58.8 + 2.9 per cent diploid) and after 10 hours' incubation (84.4 + 4.4 per cent diploid). Apparently diploid cells can resume mitosis in vitro before the less adaptable aneuploids (P = <.01). The "direct squash" of freshly aspirated marrow, therefore, provides a more accurate index of the leukemic population components existing in vivo. All published chromosome data for human and mouse leukemias are summarized, and their possible etiologic implications are discussed.

EXPANSION OF HUMAN CORD BLOOD PRIMITIVE PROGENITORS IN SERUM-FREE MEDIA USING HUMAN BONE MARROW MESENCHYMAL STEM CELLS

Ex vivo expansion of human umbilical cord blood cells (HUCBC) is explored by several investigators to enhance the repopulating potential of HUCBC. The proliferation and expansion of human hematopoietic stem cells (HSC) in ex vivo culture was examined with the goal of generating a suitable clinical protocol for expanding HSC for patient transplantation. Using primary human mesenchymal stem ...

Advances in Hematopoietic Stem Cell Mobilization and Peripheral Blood Stem Cell Transplantation

Hematopoietic stem/progenitor cells (HSPCs) which give rise to different blood cell types are present within the bone marrow microenvironment, especially in flat bones such as skull, vertebrae, pelvis and chest. Interacting factors such as stromal derived factor-1/CXCR4, very late antigen-4/vascular cell adhesion molecule-1, Lymphocyte function-associated antigen-1/ intercellular adhesion molec...

Abnormal developmental control of replication-timing domains in pediatric acute lymphoblastic leukemia.

Abnormal replication timing has been observed in cancer but no study has comprehensively evaluated this misregulation. We generated genome-wide replication-timing profiles for pediatric leukemias from 17 patients and three cell lines, as well as normal B and T cells. Nonleukemic EBV-transformed lymphoblastoid cell lines displayed highly stable replication-timing profiles that were more similar ...

Effects of 35 Hz, 2 mT Magnetic Field on Peripheral Blood Lymphocytes of Human In Vitro and Rat In Vivo

In this research we have studied the effects of extremely low frequency magnetic field on the peripheral blood lymphocytes chromosomes of human in vitro and rat in vivo. By this means, we used 60 blood samples of 10 men as well as 40 rats. Of 60 human blood samples ,40 samples (in two group of 20) were selected as test groups and placed into the 2 mT and 35 Hz magnetic field. The blood samples,...

Commentary on Sandberg et al., "The In Vivo Chromosome Constitution of Marrow".

See related article by Sandberg et al., Cancer Res 1961;21:678-89. German pathologists in the late nineteenth century appear to have been the first to publish observations of mitotic abnormalities in many types of human cancers and to suggest that they might have an important role in their pathogenesis (1). About two decades later, Theodore Boveri published his famousmonograph for which he rece...
